癌症分泌自身抗体的作用


1. 促进癌细胞恶变
癌症分泌的自身抗体具有类似于生长因子的作用,直接促进癌细胞的恶变(包括增殖、迁移、侵袭和抗凋亡)
2. 促进免疫逃逸
癌症分泌的自身抗体,通过特定的糖基化修饰(如Ans162位点的唾液化修饰),可以从促进免疫作用转变为免疫抑制作用
这些唾液酸化的IgG分子可以通过与树突状细胞上的唾液酸受体DC-SIGN结合,抑制树突状细胞(DCs)和CD4+T细胞的功能。
癌症细胞分泌抗体可以抑制ADCC功能,下调NK细胞的细胞毒活性。
3.促肿瘤炎症
在脂多糖(LPS)刺激后,癌症分泌IgG显著上调。癌症来源的IgG,可以通过TLR4/TRIF/NF-kB通路刺激肿瘤相关巨噬细胞释放IL-1b来诱导炎症。
